abstract = {An analytical transport-model was developed to simulate the propagation of a contaminant in one- and two-dimensional transient flow in groundwater. it is proved that the distribution of concentration at a given time and for a given discharge is identical to that obtained for a different discharge if the volumetric flux of water is the same in the two cases. the results of simulations have been compared with results obtained using the mt3dms numerical model. there is good agreement when the calculated concentrations are flux-weighted concentrations. on the other hand, there is a notable divergence when the resident mode is considered. resident mode concentrations express the mass per unit volume whereas flux mode concentrations express the ratio of mass flux to fluid flux. the solutions presented in this paper can thus be a useful alternative to code mt3dms when the objective is to simulate concentrations in transient flow according to a resident mode},

abstract = {Thermal spraying is often used to create coatings protecting parts against physico-chemical aggressions or improving superficial mechanical properties. It involves several problems because of the high temperatures of the process: cracks, porosity, coating adhesion. The authors use Vickers interfacial indentation test to characterize adhesive fracture between substrates and coatings. From the Lawn’s relation two curves: load/crack length ratio (P/C3/2) vs crack length (C1/2) and lnP vs lnC are drawn for varying loads. They allow the determination of the type of residual stress, a coating fracture “toughness”/residual stress ratio (KC/σr) characterizing interfacial behavior and may lead, in some cases, to residual stress σr. Influence of superficial remelting by CO2 laser of cermet coating Cr3C2/Ni–Cr deposited on NiCrAlY covered steel and of annealing of Cr3C2/Ni–Cr coated cast iron are then studied. Possibility of reducing residual stress is shown at the same time as the interest of both approaches.},

abstract = {Angiotensin-converting enzyme (ace) is a zinc metallopeptidase that plays a major role in blood homoeostasis and reproduction in mammals. in vertebrates, both transmembrane and soluble ace, containing one or two homologous active sites, have been characterized. so far, several aces from invertebrates have been cloned, but only in insects. they are soluble and display a single active site. using biochemical procedures, an ace-like activity was detected in our model, the leech, theromyzon tessulatum. annelida is the most distant phylum in which an ace activity has been observed. to gain more insight into the leech enzyme, we have developed a pcr approach to characterize its mrna. the approx. 2 kb cdna has been predicted to encode a 616-amino-acid soluble enzyme containing a single active site, named ttace (t. tessulatum ace). surprisingly, its primary sequence shows greater similarity to vertebrates than to invertebrates. stable in vitro expression of ttace in transfected chinese-hamster ovary cells revealed that the leech enzyme is a functional metalloprotease. as in mammals, this 79 kda glycosylated enzyme functions as a dipeptidyl carboxypeptidase capable of hydrolysing angiotensin i to angiotensin ii. however, a weak chloride inhibitory effect and acetylated n-acetyl-sdkp (ac sdackp) hydrolysis reveal that ttace activity resembles that of the n-domain of mammalian ace. in situ hybridization shows that its cellular distribution is restricted to epithelial midgut cells. although the precise roles and endogenous substrates of ttace remain to be identified, characterization of this ancestral peptidase will help to clarify its physiological roles in non-insect invertebrate species.},

Angiotensin-converting enzyme (ace) is a zinc metallopeptidase that plays a major role in blood homoeostasis and reproduction in mammals. in vertebrates, both transmembrane and soluble ace, containing one or two homologous active sites, have been characterized. so far, several aces from invertebrates have been cloned, but only in insects. they are soluble and display a single active site. using biochemical procedures, an ace-like activity was detected in our model, the leech, theromyzon tessulatum. annelida is the most distant phylum in which an ace activity has been observed. to gain more insight into the leech enzyme, we have developed a pcr approach to characterize its mrna. the approx. 2 kb cdna has been predicted to encode a 616-amino-acid soluble enzyme containing a single active site, named ttace (t. tessulatum ace). surprisingly, its primary sequence shows greater similarity to vertebrates than to invertebrates. stable in vitro expression of ttace in transfected chinese-hamster ovary cells revealed that the leech enzyme is a functional metalloprotease. as in mammals, this 79 kda glycosylated enzyme functions as a dipeptidyl carboxypeptidase capable of hydrolysing angiotensin i to angiotensin ii. however, a weak chloride inhibitory effect and acetylated n-acetyl-sdkp (ac sdackp) hydrolysis reveal that ttace activity resembles that of the n-domain of mammalian ace. in situ hybridization shows that its cellular distribution is restricted to epithelial midgut cells. although the precise roles and endogenous substrates of ttace remain to be identified, characterization of this ancestral peptidase will help to clarify its physiological roles in non-insect invertebrate species. |

abstract = {At the present time, there is little information on mechanisms of innate immunity in invertebrate groups other than insects, especially annelids. in the present study, we have performed a transcriptomic study of the immune response in the leech theromyzon tessulatum after bacterial challenge, by a combination of differential display rt (reverse transcriptase)–pcr and cdna microarrays. the results show relevant modulations concerning several known and unknown genes. indeed, threonine deaminase, malate dehydrogenase, cystatin b, polyadenylate-binding protein and α-tubulin-like genes are up-regulated after immunostimulation. we focused on cystatin b (stefin b), which is an inhibitor of cysteine proteinases involved in the vertebrate immune response.we have cloned the full-length cdna and named the t. tessulatum gene as tt-cysb. main structural features of cystatins were identified in the derived amino acid sequence of tt-cysb cdna; namely, a glycine residue in the n-terminus and a consensus sequence of gln-xaa-val-xaa-gly (qxvxg) corresponding to the catalytic site. moreover, tt-cysb is the first cystatin b gene characterized in invertebrates. we have determined by in situ hybridization and immunocytochemistry that tt-cysb is only expressed in large coelomic cells. in addition, this analysis confirmed that tt-cysb is up-regulated after bacterial challenge, and that increased expression occurs only in coelomic cells. these data demonstrate that the innate immune response in the leech involves a cysteine proteinase inhibitor that is not found in ecdysozoan models, such as drosophila melanogaster or caenorhabditis elegans, and so underlines the great need for information about innate immunity mechanisms in different invertebrate groups.},

abstract = {The paper deals with evaluation and improvement of two recent explicit algebraic turbulent stress models (EASMs). The first model was derived by Gatski and Rumsey (2001 Closure Strategies for Turbulent and Transitional Flows ed B E Launder and N D Sandham (Cambridge: Cambridge University Press) pp 9–46) and the second is the one devised by Wallin and Johansson (2000 J. Fluid Mech. 403 89–132). These models are studied for the turbulent flow through a square duct which involves a secondary flow and significant anisotropy between the turbulent Reynolds stress tensor components. An a priori evaluation of these models is made using direct numerical simulation (DNS) results of Navier–Stokes equations. In order to handle wall proximity effects, a damping function is suggested. The material frame-indifference (MFI) of these models is studied using the eigenvectors of the rate-of-deformation tensor and their angular velocities. This procedure allows us to evaluate an objective vorticity tensor. For this flow, it is shown that the weak-equilibrium assumption used in the derivation of EASMs is verified, i.e. the material derivative of the anisotropy tensor b is negligeable (Db/Dt ≈ 0). Comparisons of results from EASM and from DNS shows that these models are able to predict reasonably well such flows.},

abstract = {Water coning causes a reduction of oil production and an increase of production costs. Dual completion (downhole water sink) is one of the methods adopted to attenuate water coning. This work describes numerical results associated with this completion technique. The water cone shape and water breakthrough time are investigated to define the mechanism and performance of this technical procedure. The numerical results show that dual completion deforms the shape of the cone. For instance, the top of the water–oil interface is shifted away from the well yielding (under high water production rates) oil breakthrough into water perforations. The water breakthrough is proportional to dimensionless density difference and horizontal permeability and inversely proportional to oil production rate, mobility, and anisotropy ratios. High oil production rates yield elevation of water coning height that intercepts oil flow. Paradoxically, high production rate at water sink is not recommended, the improvement of water breakthrough begins when dimensionless density difference is greater than 0.05. The dual completion technique delays water breakthrough time (BT*). In general, the BT* is delayed by two times that of single completion and critical oil rate is augmented compared to single completion.},
