abstract = {The mechanism of oxidative burst induced by lead in vicia faba excised roots was investigated by luminol-dependent chemiluminescence. results showed that lead triggered a rapid and dose-dependent increase in chemiluminescence production. in this study, specific inhibitors of putative reactive oxygen species (ros) sources were used to determine the mechanism of lead-induced ros generation. this generation was sensitive to dephenylene iodonium (dpi), quinacrine and imidazole, some inhibitors of the nadph-oxidase and not inhibited by other putative ros sources inhibitors. data reported in this work clearly demonstrated the pivotal role of nadph-oxidase-like enzyme in early steps of lead-induced oxidative burst. to investigate the respective implication of calmodulin and protein kinase (pk) in leadinduced nadph-oxidase activation, excised roots were treated with the calmodulin inhibitor w7 or with the pk inhibitor staurosporine. the chemiluminescence generation inhibition by these inhibitors illustrated the role of pk in lead-induced nadph-oxidase activation and revealed a calmodulin-dependent step. using the calcium entry blocker la3+ or different concentrations of calcium in the extracellular medium, our data highlighted the implication of ca2+ channel in leadinduced oxidative burst.},

abstract = {Lead metallurgical slags are partially vitrified materials containing residual amounts of zn, pb, cr, cd and as. these hazardous materials are generally buried on heaps exposed to weathering. in this study, leaching behavior of lead blast furnace slags has been tested using pure water and open flow experiments. it appears that in such far from equilibrium and slightly acidic conditions, the main phase to be altered is the vitreous phase. as for lunar, basaltic and nuclear glasses, alkalis/proton exchanges prevail and lead to the formation of a non-protective altered layer enriched in si, fe and al. the composition of the altered layer is quite constant except for si whose concentration decreases towards the leachate interface. owing to their sizes, micrometric pb droplets are not always totally dissolved at the slag surface. nevertheless, nanometric pb droplets are instantaneously dissolved while a surrounding altered layer is formed. this leads to high pb releases in open flow systems. leachate chemistry and dissolution rates of the vitreous phase are closely comparable to previous leaching tests with basaltic and nuclear glasses in conditions far from equilibrium. moreover, this study confirms that fe is a stable element in such conditions},

abstract = {Previous studies evidenced that cystatin b-like gene is specifically expressed and induced in large circulating coelomic cells following bacterial challenge in the leech theromyzon tessulatum. in order to understand the role of that cysteine proteinase inhibitor during immune response, we investigated the existence of members of cathepsin family. we cloned a cathepsin l-like gene and studied its tissue distribution. immunohistochemical studies using anti-cathepsin l and anti-cystatin b antibodies and ultrastructural results demonstrated the presence of three distinct coelomic cell populations: (1) the chloragocytes, which were initially defined as large coelomocytes, (2) the granular amoebocytes and (3) small coelomic cells. among those cells, while chloragocytes contain cystatin b and cathepsin l, granular amoebocytes contain only cathepsin l and the third cell population contains neither cathepsin nor inhibitor. finally, results evidenced that cathepsin l immunopositive granular amoebocytes are chemoattracted to the site of injury and phagocyte bacteria},

abstract = {Primary gold deposits in burkina faso occur in paleoproterozoic birimian belt formations (2.0 ga). mineralization was synchronous with regional metamorphism and deformation, and is either hosted within, or is adjacent to, quartz-bearing veins. these are classical characteristics of epigenetic gold deposits in precambrian metamorphic terranes and permit to classify the mineralized sites from burkina faso as orogenic-type gold deposits. a review of data collected over the past decade by our team permits to recognize two main styles of gold mineralization: (1) quartz-vein hosted; this style occurs in all lithologies, the veins are deformed and gold is principally concentrated within the veins, associated with either sulfides or tourmaline. (2) disseminated; this style occurs exclusively in albitites (and to a lesser extent listvenites) with gold occurring mainly within alteration halos of generally undeformed quartz-albite-carbonate vein. quartz-vein and disseminated styles of mineralization can be associated within the same deposit. albitites and listvenites are alteration products of mainly calc-alkaline igneous rocks of felsic to ultramafic composition, respectively. the predominant alteration assemblage consists of chlorite, albite, carbonate, and pyrite. sulfides occur as fine masses commonly in the alteration halos close to vein margins and consist mainly of pyrite and arsenopyrite, depending on host-rock composition. gold occurs as free native metal and, locally, in form of tellurides, in fissures or as inclusions within pyrite and arsenopyrite. two main populations of fluid inclusions are associated with the gold deposits, independently of the mineralization style: (1) carbonic inclusions consisting of up to 90 mol% co2 (plus n2 and ch4) and (2) aqueous-carbonic fluid inclusions with moderate salinities. interestingly, the disseminated gold style deposits of burkina faso, which have the highest economic potential, show strong similarities with the world-class ashanti deposit, in neighboring ghana.},

abstract = {The aim of this in situ study was to investigate the fatty acid (fa) composition and content in roots and shoots of lolium perenne and trifolium repens, grown under heavy metal stress (cd, pb, zn). the composition of fa was quite similar for the two plants and the two organs; main fa were palmitic acid (c16:0), oleic acid (c18:1), linoleic acid (c18:2) and linolenic acid (c18:3). for both plants, the major fa that characterized the roots was c18:2 whereas c18:3 was the prominent fa in shoots. for the first sampling (s1), in the roots of l. perenne and t. repens, polyunsaturated fatty acids (pufa) were affected by metal contamination while, in the second sampling (s2), pufa were affected in the shoots of the two plants. this alteration of pufa was well correlated with the bioaccumulation factor of metals which decreased in roots and increased in shoots with the time. moreover, a positive correlation was found between the pufa decrease and the malondialdehyde (mda) content, indicating the occurrence of a lipid peroxidation induced by the metal stress},

abstract = {The aim of this work was to identify in eisenia fetida genes whose expression are regulated following exposure to a complex mixture of metallic trace elements (mte) representative of a highly polluted smelter soil. suppression subtractive hybridization (ssh) was used to construct cdna libraries enriched in up- or down-regulated transcripts in the immunecirculating cells of the coelomic cavities, namely coelomocytes, from worms exposed to metallic pollution. among 1536 ssh-derived cdna clones sequenced, we identified 764 unique ests of which we selected 18 candidates on the basis of their redundancy. these selected candidates were subjected to a two-step validation procedure based on the study of their expression level by real-time pcr. the first step consisted in measuring the expression of the 18 candidates in worms exposed to artificial contaminated soil. the second step consisted in measuring the expression in animals exposed to a ‘‘naturally’’ contaminated soil sampled close to a smelter. both steps allowed us to highlight 3 candidates that are strongly induced in worms exposed to a smelter polluted soil. these candidates are: the well-known mte-induced cd–metallothionein and 2 original biomarkers, lysenin, and a transcript, which cloning of the complete coding sequence identified as the coactosin-like protein.},

abstract = {Metallothioneins (mts) are central to trace metal homeostasis and detoxification throughout biological systems. prokaryotes, plants, and fungi utilize both gene encoded cysteine-rich polypeptides (classically designated class i and ii mts) and enzymatically synthesized cysteinerich peptides (classically designated class iii mts or phytochelatins). in contrast, although gene encoded mts are ubiquitous in animal species the identification of a functional phytochelatin synthase in the nematode caenorhabditis elegans, a representative member of the ecdysozoa, provided the first evidence for these metal-binding peptides in animals. by exploiting the conservation observed between species we have been able to clone and transcriptionally characterize a phytochelatin synthase from the immune cells of the earthworm eisenia fetida, the first evidence for its presence in a phylum belonging to the lophototrochozoa. the complete coding sequence of this enzyme was determined and the phylogenetic relationship to plant, yeast and nematode enzymes elucidated. temporal- and dose-profiling of the transcriptional regulation of phytochelatin synthase and mt in response to cadmium was performed by using real-time pcr.},

abstract = {An analytical transport-model was developed to simulate the propagation of a contaminant in one- and two-dimensional transient flow in groundwater. it is proved that the distribution of concentration at a given time and for a given discharge is identical to that obtained for a different discharge if the volumetric flux of water is the same in the two cases. the results of simulations have been compared with results obtained using the mt3dms numerical model. there is good agreement when the calculated concentrations are flux-weighted concentrations. on the other hand, there is a notable divergence when the resident mode is considered. resident mode concentrations express the mass per unit volume whereas flux mode concentrations express the ratio of mass flux to fluid flux. the solutions presented in this paper can thus be a useful alternative to code mt3dms when the objective is to simulate concentrations in transient flow according to a resident mode},

abstract = {A cdna sequence with homologies to members of the lps-binding protein and bactericidal/permeability-increasing protein (bpi) family was identified in the oyster crassostrea gigas. the recombinant protein was found to bind lps, to display bactericidal activity against escherichia coli, and to increase the permeability of the bacterial cytoplasmic membrane. this indicated that it is a bpi rather than an lps-binding protein. by in situ hybridization, the expression of the c. gigas bpi (cg-bpi) was found to be induced in hemocytes after oyster bacterial challenge and to be constitutive in various epithelia of unchallenged oysters. thus, cg-bpi transcripts were detected in the epithelial cells of tissues/organs in contact with the external environment (mantle, gills, digestive tract, digestive gland diverticula, and gonad follicles). therefore, cg-bpi, whose expression profile and biological properties are reminiscent of mammalian bpis, may provide a first line of defense against potential bacterial invasion. to our knowledge, this is the first characterization of a bpi in an invertebrate.},
